Nicotinamide NAF is an over-the-counter medicine used against pellagra (vitamin B3 deficiency), both therapeutically and prophylactically.
Pellagra is a syndrome caused by a lack of nicotinamide (vitamin B3). Typical symptoms of pellagra are loss of appetite, fatigue, anemia and skin changes, such as dermatitis and stomatitis, hyperkeratinization and hyperpigmentation, magenta tongue (diffuse red tongue, which is due to inflammation). Other symptoms can be diarrhoea, abdominal pain, headache and mental and neurological disturbances. Untreated pellagra is fatal due to heart failure.
Nicotinamide forms in the body together with adenine nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP), coenzymes that play a very important role in oxygen metabolism in the body.
Both nicotinamide and nicotinic acid can be used as vitamin B3 supplements, but nicotinamide is preferred, both because it is formed naturally in the body and because nicotinic acid has a number of side effects. Nicotinic acid must also be converted to nicotinamide before it can be used in nucleotide metabolism.
